제목: Unraveling Brain Development in Health, Disease, and Evolution Through ASPM


아래는 이번 세미나의 Abstract 입니다.

The cerebral cortex or neocortex—our brain's outermost and largest structure—underpins complex cognitive and social functions. Its development is tightly orchestrated, and disruptions lead to neurodevelopmental disorders such as autism and schizophrenia. Evolutionarily, the cortex has undergone remarkable expansion in the last 2-3 million years. At the University of Connecticut School of Medicine, the Bae lab investigate the development, disease, and evolution of the cerebral cortex using ASPM (abnormal spindle-like microcephaly-associated) as a molecular entry point. ASPM encodes an evolutionarily divergent centrosomal protein essential for the proliferation of cortical progenitors, particularly ventricular/apical and outer/basal radial glia. Our research focuses on two key directions. (1) ASPM loss-of-function mutations cause microcephaly (‘small head’), with species-specific severity. Using ASPM knockout ferrets, mice, and human cortical organoids, we aim to uncover why large, folded brains are more vulnerable to ASPM loss. (2) We recently identified the first ASPM gain-of-function mutation associated with autism, which leads to macrocephaly (‘large head’) and social-cognitive deficits in mice. Through collaboration, we are investigating how excessive embryonic neurogenesis alone can drive specific behavioral phenotypes. In this seminar, I will share our recent findings and their implications for understanding cortical development in evolution and disease.
